Abstract: Pancreatic lipase plays an important role in converting triglyceride into long-chain fatty acids and glycerol, then becomes a therapeutic target for obesity treatment. Quercetin and chrysin are able to inhibit pancreatic lipase, which is potentially developed for obesity treatment. This study aimed to identify quercetin and chrysin derived from the methanol extract of raja and kepok banana peels using High Performance Liquid Chromatography (HPLC) and Liquid Chromatography Mass Spectrometer (LC-MS) for the development of obesity treatment. Raja and kepok banana peels were purchased from a fruit seller in Klaten City. Raja and kepok banana peels were extracted using the maceration method with 80% (v/v) methanol solvent. Quercetin and chrysin compounds were identified using HPLC and LC-MS at the Integrated Laboratory UNS and Universitas Diponegoro, respectively. Quercetin concentration was calculated using linear regression of diluted standard quercetin. The quercetin concentration in the raja peel extract was 0.338 mg/dL but was not detected in the kepok banana peel extract. The peak of chrysin in raja and kepok banana peel extract was not detected. LC-MS analysis identified chrysin in raja banana peel extract. In conclusion, raja banana peel extract is a potential source of quercetin and chrysin for alternative natural lipase inhibitors for obesity treatment. Further research is required carry out extraction using other solvents, isolate and purify quercetin and chrysin from raja banana peel extract, which will be used for in vitro and in vivo studies.

Abstract: This study investigates the effectiveness of adding carboxymethyl chitosan during steam cooking as an adsorbent for detoxifying paralytic shellfish poisoning toxins (PSP toxins) in the scallop Chlamys nobilis. Toxin analysis using a mouse bioassay test and hydrophilic interaction liquid chromatography–tandem mass spectrometry method (LC–MS) showed that most of the PSP toxins (>80%) were contained in the visceral compartment of the raw scallops. Overall, 82.2Mu/100g of PSP toxins were released from scallop tissues during steaming. The toxicity of the soup steamed with 0.1%, 0.3%, 0.6%, 1.0% and 1.5% carboxymethyl chitosan decreased the toxin content by 35.6%, 44%, 48.4%, 53.3% and 56.9% (p<0.005), respectively. The relative PSP toxin content in the raw adductor muscle was comparable to those after steaming or steaming with carboxymethyl chitosan (p > 0.05).The PSP toxin concentrations in the adductor muscles, gills + mantle and visceral compartments steamed with 0.1%, 0.3%, 0.6%, 1% and 1.5% carboxymethyl chitosan were not significantly different from those in the corresponding raw samples. The LC–MS analysis showed that the adsorbance of carboxymethyl chitosan for each of the PSP toxins was positive correlation with concentration, although the uptake efficiency of each toxin was different. The reduction in toxin content of all analyzed toxins reached 45.6%, 30.2%, 44.5%, 55.9% and 37.6% under the corresponding carboxymethyl chitosan concentrationsis.

Abstract: With the rapid increasing happening frequency of red tide in China, the urgent pollution issue of paralytic shellfish poisoning (PSP) toxins in aquatic products is becoming more serious and attacking greater attention. Thus, the routine monitoring of PSP toxins in aquatic products need to be vigorously enforced. In this study, the LC-MS-MS method for eight PSP toxins was developed and applied for the quantitative analysis in sea foods from different areas of China. The result showed the proposed method can completely meet the need for their routine monitoring purpose.

Abstract: The molecule weight and distribution of molecule weight of a kind of medium temperature phenol formaldehyde resin(MTPF)was detected through Liquid Chromatogram-mass spectrum instrument(LC-MS).The result showed that the distribution scope of molecule weight of the MTPF during the initial stage was approximately 200-400,and the degree of polymerization of the MTPF molecule was 2-4,At the same time,the MTPF during initial stage still has a certain amount of medium outcome hydroxymethyl phenol which was formed through the addition reaction between phenol and formaldehyde.The detection result,from the MTPF during initial stage through ZEN3600 nanometer grain analysis instrument, proved that the grain size of the MTPF during initial stage was about 80-180nm,and the distribution of the grain size presented a normal one that the grain size 96-130nm was about 67%.
