Pyridine-3,5-dicarbonitriles (PD) of general structure 1 demonstrate activity against a wide range of biological targets[13-17].
The typical multi-component reaction (MCR) employing aldehydes, malononitrile and thiols to synthesize structure 1 proceeded only in low to moderate yield (route A in Scheme 1)[18-19].
By studying the reaction mechanism, especially the oxidation mechanism of the intermediate 1,4-dihydropyridine 2, we improved the reaction yield by about 50% via using an extra equivalent of the Knoevenagel adduct 3 in situ (route B in Scheme 1), concluding that the structure 3 played a significant role as oxidant in the final oxidation step[20].
Therefore, the optimal temperature for this MCR would be 90 ºC for 30 seconds in ethanol using MWI, representing a significant move toward green chemistry by substantial savings on energy consumption.
