However, the first thymidine kinase catalyzed phosphorylation of d4T to its monophosphate is the limiting step in cells because of d4T is different from the natural nucleosides on the structure. [4, 5] A variety of phospholipid derivatives of d4T have been synthesized to overcome the dependence on nucleoside kinase activation.
