Abstract:

Inosine 5′-triphosphate (ITP) and 5′-diphosphates (IDP) were efficiently synthesized. Phosphitylation, acid-catalyzed hydrolysis, and oxidative coupling with piperidine afforded inosine phosphoropiperidate benzyl ester intermediate in good yield. Inosine polyphosphates were obtained by coupling the deprotected phosphoropiperidate with 4,5-dicyanoimidazole (DCI) as the activator.
