Abstract:

A novel and efficient method for the preparation of P1-stavudine-P2-acyclovir diphosphate has been developed. Acyclovir phosphoropiperidate was prepared from a redox condensation with 2,2′-dithiodianiline/PPh3. Then, the coupling of phosphoropiperidate with d4T phosphate in the presence of 4,5-dicyanoimidazole as the activator afforded the desired asymmetrical dinucleoside diphosphate in high yield.
