Optimization of NDV Encapsulated in N-2-HFCC/CMC Nanoparticles Preparation Conditions

Abstract:

The weak solubility of chitosan is the main disadvantage as a delivery carrier for drug application. In this study, a novel soluble in water chitosan derivative N-2-hydroxypropyl dimethyl ethyl ammonium chloride chitosan (N-2-HFCC) was successfully synthesized with low toxicity, a high safety level and used to make nanoparticles as a delivery vehicle for live attenuated Newcastle disease vaccine. In addition, Newcastle disease virus (NDV) La Sota encapsulated in the N-2-HFCC/CMC nanoparticles (NDV/La Sota-N-2-HFCC/CMC-NPs) was prepared by a polyelectrolyte complexation method. The preparation technology of NDV/La Sota-N-2-HFCC/CMC-NPs was optimized by single factor and orthogonal test. The results of this study demonstrated that the optimal conditions for the preparation of NDV/La Sota-N-2-HFCC/CMC-NPs is stirring speed and stirring time of 1200 r/min and 40 min, NDV dosage of 2.0 ml, the N-2-HFCC concentration of 1.25 mg/ml and the CMC concentration of 2.0 mg/ml.
