Abstract: Poly (ethyleneimine) (PEI) is utilized as the delivery vector for vascular endothelial growth factor (VEGF) 165-angiopoietin-1 (Ang-1) dual gene simultaneous expression plasmid. The influences of PEI/pDNA ratios on the packing effect, cytotoxicity and transfection efficiency are investigated. The result of agarose gel electrophoresis suggesst that pDNA are entrapped into PEI completely when N/P ratio exceeds 3/1. MTT assay demonstrates that the cell viability is over 90% when the PEI/pDNA (w/w) ratios is respectively 1/2, 2/2, 3/3, 4/2 or 5/2. The L929 cells are transfected with PEI/pDNA in vitro, the results show that the fluorescence intensity and transfection efficiency could reach their highest levels when the PEI/pDNA ratio is 3/2. In general, this study provides a novel method for future in vivo transfection investigations.

Abstract: A series of bicyclic furo[2,3-d]pyrimidine nucleosides conjugated with N-diisopropylphosphoryl amino acids were synthesized from 2′-deoxy-5-iodouridine. The key step is its cross coupling with the 5-phthalimido-1-pentyne under Sonogashira reaction condition. 5-Endo-dig electrophilic cyclization catalyzed by AgNO3 is used to afford the furo[2,3-d]pyrimidines.

Abstract: Objective: To explore the expression of EGFR and PCNA in lung squamous cell carcinomas and lung adenocarcinomas, and the relationships between their overexpression and cancer tissue typing or lymphatic metastasis. Method: Detect the expression of EGFR and PCNA in 44 cases of similarly differentiated cancer tissue and 16 cases of normal lung tissue by immunohistochemistry. Result: The expression of EGFR and PCNA in lung cancer tissue was significantly higher than that in normal lung tissue. Meanwhile, no significant difference existed between expression of PCNA and EGFR in lung squamous carcinomas and that in lung adenocarcinomas (P>0.05), nor between lymph node metastatic tissue and negative lymph node metastatic tissue (P>0.05). Conclusion: EGFR and PCNA were involved in the onset and development of lung cancer, and had some correlation with lung cancer. No significant correlations were observed between EGFR or PCNA overexpression and lymph node metastasis.

Abstract: A series of carbohydrate-conjugated 4-methylumbelliferone were synthesized using 1,3-dipolar cycloaddition “click chemistry” of the azide and alkyne. The reaction is straight forward and in high yield. The strategy will be useful for synthesizing glycoconjugated derivatives of natural products.

Abstract: Eight compounds were isolated from the ethanol extract of G. algida Pall. the structure of these compounds were identified as: oleic acid (1), linoleic acid (2), octacosanoic acid (3), β-sitosterol (4), 2,3-dihydroxybenzoic acid ethyl ester (5), n-butyl isobutyl phthalate (6), daucosterol (7), 5,8-dimethoxy furocoumarin (8). These structures were identified by NMR MS IR and other spectroscopic techniques. These compounds were isolated from this plant for the first time.

Abstract: The current study was designed to investigate the anti-fatigue effects of polysaccharide of Gynostemma pentaphyllum Makino (GMP). The mice were randomly divided into four groups: control group, low-dose GMP-treated group, middle-dose GMP-treated group and high-dose GMP-treated group. The animals of control group received an oral administration of physiological saline in a volume of 1.0 mL, and the animals of treated group received the same volume of GMP (50, 100 and 200 mg/kg bodyweight/day) for 28 days. After 28 days, anti-fatigue effects of GMP were assessed by forced swimming test and some biochemical parameters related to fatigue including blood lactic acid (BLA), blood urea nitrogen (BUN), superoxide dismutase (SOD), glutathione peroxidase (GPH-Px), liver glycogen and muscle glycogen were measured using commercial kits. The results demonstrated that GMP prolonged the exhaustive swimming time, increased the liver glycogen and muscle glycogen contents, and decreased the BLA and BUN levels. It also improved the SOD and GPH-Px activities in blood of mice. These results indicated that FPR had anti-fatigue effects.
