Abstract: Sericin peptides have a high proportion of hydrophilic amino acid residues, which are considered to protect cells from freezing stress due to the maintenance of water in the cells. In this paper, the hypothermia protection effect of sericin peptides was studied by reading optical density of cells at 600 nm, which was detailedly optimized to be a direct and practical spectrophotometric method. The survival rate of Lactobacillus bulgaricus after frozen storaged with addition of 0.1 mg/mL sericin peptides was close to that of 10% (v/v) glycerol (p > 0.05). The survival rate of 5 mg/mL sericin peptides was up to 64.17%.

Abstract: Pinus armandi franch is a unique specialty plant in China and its seed oil contains high levels of essential fatty acids (EFA), particularly linoleic acid (LA), which has several pharmaceutical properties. Pinus armandi franch seed oil is a nice resource of linoleic acid with a content of 63% of the total fatty acids. Linoleic acid is an essential fatty acid, whose absence in a normal diet is responsible for the development of various abnormal disorders. This work reported purified LA from Pinus armandi franch seed oil could lower MDA content of lipid peroxidation on rats with hyperlipidemia significantly. TAC activity of liver, heart and serum was enhanced significantly, as well as SOD activity was increased. It demonstrated purified LA from Pinus armandi franch seed oil could improve antioxidant levels of hyperlipidemia rats effectively, enhance the activity of antioxidant enzyme and reduce the content of lipid peroxide, thereby improving lipid metabolism.

Abstract: A Box-Behnken experimental design and response surface methodology (RSM) was employed to optimize the parameters of ultrasonic extraction of phenolics from longan seed based on the single-factor experiments. The effects of extraction temperature, methanol concentration and extraction time on the extraction yield of phenolics from longan seed were investigated. The major parameter influencing the extraction was the extraction temperature. In this experiment, the maximal extraction yield of phenolics was obtained with 5.931 ± 0.016% (n = 3) at extraction temperature 82°C, methanol concentration 50% and extraction time 41 min. Under optimal conditions, the activity predicted by the model agreed well with experimental data.

Abstract: Hemagglutination titer of crude lectin from four traditional chinese medicine was detected. Lectins from Radix Polygalae and Saxifraga stolonifera agglutinated red blood cells of rabbit, rat and chook potently. Heteropappus hispidus (Thunb.) Less lectin also agglutinated the three kinds of red blood cells, but hemagglutination titer was lower. Hemagglutination effect of Lepisorus waltonii (Ching) Ching lectin only appeared in rat red blood cells.

Abstract: Objective To evaluate the effectiveness of prokaryotic expression of fusion proteins pp150-pp65 of human cytomegalovirus (hCMV) for its application as antigen, the fusion protein of pp150-pp65 were expressed in prokaryotic expression system and purified by Ni-NTA affinity chromatography column for preparing the colloidal gold kit. Methods Using DNA from HCMV strain as template, the genes encoding pp150 and pp65 protein fragment were amplified by PCR technique, respectively. After confirmed by DNA sequence analysis, the recombinant plasmid pET28a-pp150-pp65 was transformed into E.Coil.BL21(DE3) and induced to express with IPTG. The expressed fusion protein was characterized by SDS-PAGE and western blot after purified, then we used the purified fusion protein to develop combined detection kit of IgM/IgG antibody against HCMV (colloidal gold method) with Beijing Innovita Bio-tech Co., Ltd for detecting the samples, compared with the imported kits (ELISA). Results The gene of fusion fragment pp150-pp65 was correctly amplified and the recombinant vector was successfully constructed. The purified protein with the molecular weight of 45KD had good antigenicity by western blot. The protein was subjected to assay with an ELI SA capture kit in its specific and sensitive assay based on colloidal gold nanoparticles, testing of 600 serum samples indicated that this kit had a sensitivity of 92.7%;, specificity of 83.1%, crude consistency o f 90.2%, compared to the imported HCMV-IgG kit; the sensitivity of the kit was 88.1%, specificity was 89.2%, coarse consistency was 88.5%, compared to the imported HCMV-IgM kit; Conclusion In this experiment, the HCMV antigen with high purity and specificity (pp150-pp65 recombinant protein) was prepared effectively through genetic engineering technology. Compared to imported reagents, the colloidal gold kit consisting of fusion protein in a capture assay had high sensitivity and specificity. Preliminary clinical use warrants further development and use of this kit. Furthermore, it provides a technological basis for detection of HCMV in different stages of clinical infection.

Abstract: Juvenile turbot (Scophthalmus maximus) were exposed to acaricide fenpyroximate, and the subsequent histological, enzyme activity changes of liver tissue were examined. Histopathological examinations showed that the turbot liver is the target organ for the acaricide fenpyroximate. Analysis of liver antioxidant enzymes, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPX), in the exposed turbots also showed similar changes. These changes indicated that fenpyroximate caused the oxidative stress on the turbot liver, which induced the damage of antioxidant enzymes system and possible interruption of the mitochondrial redox respiratory chain. This could be one toxic mechanism of fenpyroximate to the juvenile turbot.

Abstract: There is increasing evidence indicating that oxidative stress plays an important role in the pathogenesis of rhabdomyolysis-induced myoglobinuric acute renal failure (ARF). In this study, protective effects of L-citrulline on glycerol-induced acute renal failure (ARF) in rats were investigated. Six groups of rats were employed in this study, after seven days of glycerol injections, the blood samples and kiney tissues were harvested for future biochemical and pathology analysis. The levels of creatinine (Cr) and urea nitrogen (BUN) in plasma, the content of malondialdehyde (MDA), glutathione (GSH), nitric oxide (NO), the activity of total nitric oxide synthase (TNOS), inducible nitric oxide synthase (iNOS), endothelial NO synthase (eNOS) and superoxide dismutase (SOD) were evaluated in kiney tissues. Consequently, treatment with L-citrulline improved an impaired intrarenal oxygenation and kidney function compare with the glycerol group, and prevented the renal oxidative stress damage as well as severe functional and morphological renal deterioration.

Abstract: Response surface methodology (RSM) and single factor test were used to optimize preparation technology on medicated serum of Yinqiaotiangan (YQTG) in mice. The influent factors of preparing medicated serum include blood sampling time, administration times, and administration dosage. The content of chlorogenic acid in medicated serum was detected by high performance liquid chromatography (HPLC). Results showed that the optimum preparation technology of medicated serum is 33 minutes of blood sampling time, 4 times of administration times and 0.024g/mL of administration concentration. Under this condition, the concentration of chlorogenic acid reached peak 33.34 μg/mL in mice medicated serum. It is seen that the RSM method effectively and accurately optimize preparation technology on medicated serum of YQTG preparation.

Abstract: This paper reported a method of amplifying ovarian biomarkers concentration. Ultrasound device radiated to ovarian cancer cell to amplify the concentration of the biomarkers in the blood, achieving early diagnosis of ovarian cancer. The device used digital frequency generator technique and adaptive load technology to produce frequency(1MHz), duty cycle (50%), power(0.3W/cm2). This study used the device to produce ultrasound exposure on vitro ovariancancer cell and determined the concentration of CA125 and CA19-9 in the blood after ultrasound exposure. The results show that the device can produce stable frequency and ultrasound power. The device radiated vitro ovarian cancer cell 5min, the biomarker CA125 concentration amplified 2.02 fold and CA19-9 concentration amplified 4.21 fold compare with no radiation ovarian cancer cell. This project developed a device for amplifying ovarian cancer biomarkers concentration which can amplify two or more biomarkers concentration simultaneously.

Abstract: The aim of this study was to evaluate the value index of tumor biomarkers for diagnosis and prediction of bone metastasis by combining pyridinoline cross-linked carboxy-terminal telopeptide of type I collagen (ICTP) in serum with other biochemical markers in patients with several cancers. All of 1253 patients and biomarkers data with cancers and the related benign diseases and 312 healthy controls were included in the study. Bioinformatics, data mining technology and statistics were carried out to calculate sum of direct positive index (SDPI) and sum of indirect positive index (SIPI). The average ICTP was higher in patients with bone metastasis from liver, gastric, lung, colorectal and pancreatic cancer, than in cancer patients without bone metastasis. For those cancers patients with bone metastasis, which could not be differentiated by single ICTP, combining with SDPI and SIPI would distinguish types of carcinama in situ from similar symptoms. The measurement of multiple tumor markers was very useful in complementary diagnosis and prediction of different carcinoma with bone metastasis bottom.
